The Study of Human Embryonic Stem Cell Culture According to Component of Culture Media in Vitro

Abstract

Over the past several years, the number of researchers culturing human embryonic stem(hES) cells have increased dramatically. One of the most important parameters surrounding ES cell culture is the maintenance of ES cells in an undifferentiated state. Therefore, many chemical reagent have been indroduced for hES cell culture. In our studies, we compared effect on undifferentiated hES cell culture in 4 of types culture medium. We cultured hES with KO-KMEM and DMEM-F12 as basic medium, and added FBS or SR to each basic medium(KO-DMEM/FBS, KO-DMEM/SR, DMEM-F12/FBS and DMEM-F12/SR). As results, we have demonstrated that the culture medium SR more efficient to maintain undifferentiated hES cell culture than FBS. But there is no significant difference between basic medium.
